摘要:
the intracellular secondary messenger cgmp mediates signals by activating two types of cgmp-dependent protein kinases (pkg) (i and ii) differentially expressed in different cells. it is generally accepted that cgmp and cgmp-dependent protein kinase (pkg) play an inhibitory role in platelet activation. however, we have recently shown that cgmp plays biphasic roles in platelet activation: at low concentrations, cgmp stimulates platelet activation, and at high concentrations it inhibits platelet activation. physiological platelet agonists induced increase of intracellular cgmp levels. a specific inhibitor of phosphodiesterase (pde) 5, viagra, enhanced intra-platelet cgmp concentration and promoted platelet activation. we further show that the stimulatory effect of cgmp is mediated by pkg i and ii.

cancers cause the deaths of millions of people annually, and seriously affect the lives of a lot more others. immunotherapy has emerged as one of the novel treatments in addition to traditional chemotherapy, surgical operation, and radiation therapy. dna vaccines have been proven to be effective and feasible in cancer patients. convenient and economic generation and modification of dna constructs, prolonged expression of whole antigenic determinants of tumor antigens of interest and costimulatory molecules make dna vaccine an attractive approach to be further explored and pushed for general clinical applications in cancer patients in the near future.

摘要:
abnormal hype angiogenesis has been found in many malignant tumors. traditionally, vascular endothelial cells (ecs) have been considered to be key cells in tumor angiogenesis. recent researches of my team and other laboratories have revealed that tumor angiogenesis at early stage of malignant tumor growth is not mainly dependent on ecs; certain tumor cells and pre-cancer stem cells may form blood vessels. using our newly-established in vitro tumor angiogenic model, we have recently demonstrated that t2-tryptophanyl trna synthetase (t2-trprs) can specially inhibit tumor angiogenesis and found that pre-cancer stem cells abnormally express ve-cadherin and form vessel-like tubes.we have further identified a motif in ve-cadherin as the binding site of t2-trprs, unraveling the mechanism of specific anti-tumor angiogenesis of t2-trprs and revealing this motif in ve-cadherin as a new target for development of novel anti-tumor angiogenic drugs.
